摘要: | 目的:研究鸡冠花正丁醇萃取物对大鼠功能失调性子宫出血的改善作用,并探讨其作用机制。方法:将60只受孕SD大鼠随机分为空白组、模型组、宫血宁胶囊组(阳性对照,0.07 g/kg)和鸡冠花正丁醇萃取物高、中、低剂量组(4.32、2.16、1.08 g/kg),每组10只。除空白组外,其余各组大鼠均在妊娠第7天ig米非司酮溶液和米索前列醇溶液造成不完全流产,以复制功能失调性子宫出血模型。从妊娠第8天开始,给药组大鼠ig相应药物,空白组和模型组大鼠ig等体积生理盐水,每天早、晚各给药1次,连续7 d。给药结束后,测定大鼠子宫出血量以及血清中血栓素(TXA2)、前列环素(PGI2)和肿瘤坏死因子α(TNF-α)含量。结果:与空白组比较,模型组大鼠子宫出血量显著增加(P<0.01),血清中TXA2含量显著减少、PGI2和TNF-α含量显著增加(P<0.01)。与模型组比较,各给药组大鼠的子宫出血量均显著减少、血清中TXA2含量均显著增加(P<0.01);宫血宁胶囊组和鸡冠花正丁醇萃取物高剂量组大鼠血清中PGI2、TNF-α含量,以及鸡冠花正丁醇萃取物中剂量组大鼠血清中TNF-α含量均显著减少(P<0.01)。结论:鸡冠花正丁醇萃取物对药物流产不全致大鼠功能失调性子宫出血有明显的改善作用,其作用机制可能与调节TXA2/PGI2动态平衡、抑制TNF-α的过度分泌有关。 |
ABSTRACT: | OBJECTIVE: To study the improvement effect of Celosia cristata n-butanol extracts on dysfunctional uterine bleeding of rats, and explore its mechanism. METHODS: 60 pregnant SD rats were randomly divided into blank group, model group, Gongxuening capsule group (positive control, 0.07 g/kg) and C. cristata n-butanol extracts high-dose, medium-dose, low-dose groups (4.32, 2.16,1.08 g/kg), 10 in each group. Except for the blank group, rats in other groups were intragastrically given mifepristone and misoprostol on 7th of pregnancy for resulting incomplete abortion to induce models of dysfunctional uterine bleeding. Then rats in administration groups were intragastrically given relevant medicines, rats in blank group and model group were intragastrically given normal saline once every morning and evening, for 7 d. On 8th d of pregnancy, uterine bleeding amount, and thromboxane (TXA2), prostacyclin (PGI2) and tumor necrosis factor α (TNF-α) contents in serum were determined. RESULTS: Compared with blank group, uterine bleeding amount in model group was significantly increased (P<0.01), TXA2 content in serum was significantly reduced, PGI2 and TNF-α contents were significantly increased(P<0.01). Compared with model group, uterine bleeding amounts in administration groups were significantly reduced, TXA2 content in serum was significantly increased (P<0.01); PGI2 and TNF-α contents in serum in Gongxuening capsule group and C. cristata n-butanol extracts high-dose group and TNF-α content in serum in C. cristata n-butanol extracts medium-dose group were significantly reduced (P<0.01). CONCLUSIONS: C. cristata n-butanol extracts show obvious improvement effect on incomplete drug abortion-induced dysfunctional uterine bleeding of rats, and the mechanism may be related to the regulation of TXA2/PGI2 dynamic balance and inhibition of TNF-α transient secretion. |
