摘要: | 目的:建立透骨香药材的质量标准。方法:采用薄层色谱法(TLC)对药材进行定性鉴别;测定药材水分、总灰分和酸不溶性灰分;采用高效液相色谱法测定药材中水杨酸甲酯的含量:色谱柱为Eclipse XDB-C18,流动相为甲醇-水(62 ∶ 38,V/V),流速为1.0 mL/min,检测波长为307 nm,柱温为30 ℃,进样量为10 μL。结果:药材的TLC图斑点清晰,分离度好。药材水分为8.2%~10.8%,总灰分为0.9%~4.0%,酸不溶性灰分为0.1%~0.9%。水杨酸甲酯检测进样量线性范围为0.045~0.73 μg(r=0.999 9);精密度、稳定性、重复性试验的RSD≤1.0%;水杨酸甲酯加样回收率为97.8%~104.3%(RSD=2.6%,n=9)。结论:该研究所建标准可用于透骨香药材的质量控制。 |
ABSTRACT: | OBJECTIVE: To establish the quality standard for Gaultheria yunnanensis. METHODS: TLC was adopted for qualitative identification of samples. Moisture, total ash and acid-insoluble ash were determined. HPLC method was used to determine the content of methyl salicylate. The determination was performed on Eclipse XDB-C18 column with mobile phase consisted of methanol-water(62 ∶ 38,V/V)at the flow rate of 1.0 mL/min. The detection wavelength was set at 307 nm, and column temperature was 30 ℃. The sample size was 10 μL. RESULTS: TLC spots of samples were clear and well separated. The moisture was 8.2%-10.8%, total ash was 0.9%-4.0% and acid-insoluble ash was 0.1%-0.9%. The linear range of methyl salicylate were 0.045-0.73 μg (r=0.999 9). RSDs of precision, stability and reproducibility tests were no more than 1.0%. The recoveries of methyl salicylate were 97.8%-104.3% (RSD=2.6%,n=9). CONCLUSIONS: The established standard can be used for quality control of G. yunnanensis. |
