摘要: | 目的:建立同时测定余甘子中5种成分含量的方法,同时对广西不同产地余甘子中5种多元酚类成分的含量进行主成分分析和聚类分析。方法:采用高效液相色谱法测定余甘子中没食子酸、没食子儿茶素、柯里拉京、诃子联苯酸、鞣花酸的含量。色谱柱为Agilent Eclipse XDB,流动相为乙腈-0.1%磷酸水溶液(梯度洗脱),流速为1.0 mL/min,检测波长为220 nm,柱温为25 ℃,进样量为20 μL。对含量测定结果进行主成分分析和聚类分析。结果:没食子酸、没食子儿茶素、柯里拉京、诃子联苯酸、鞣花酸检测质量浓度线性范围分别为1.994~31.90、0.159 8~2.556、4.533~45.33、14.75~88.49、2.956~47.30 μg/mL(r均≥0.999 8);检测限分别为0.025 6、0.027 1、0.052 9、0.186 7、0.133 1 μg/mL,定量限分别为0.085 1、0.089 3、0.170 6、0.615 2、0.441 9 μg/mL;精密度、稳定性(24 h)、重复性试验的RSD均<2.0%(n=6或n=7);加样回收率为96.19%~101.49%,RSD为0.98%~1.41%(n=6)。12批样品有3个主成分,可聚为2类,其中广西玉林市容县的余甘子综合主成分得分最高,药材质量较好。结论:建立的方法操作简便、结果准确可靠,可用于余甘子中没食子酸、没食子儿茶素、柯里拉京、诃子联苯酸、鞣花酸含量的同时测定;广西不同产地余甘子质量有差异。 |
ABSTRACT: | OBJECTIVE: To establish a method for simultaneous determination of 5 constituents in Phyllanthus emblica, and to conduct principal component analysis and cluster analysis of contents of 5 polyphenolic constituents in P. emblica from different producing areas of Guangxi. METHODS: HPLC method was adopted to determine the contents of gallic acid, epigallocatechin, corilagin, terminalia biphenyl acid and ellagic acid in P. emblica. The determination was performed on Agilent Eclipse XDB column with mobile phase consisted of acetonitrile-0.1% phosphoric acid aqueous solution (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 220 nm, and column temperature was 25 ℃. The sample size was 20 μL. The results of content determination was processed by principal component analysis and cluster analysis. RESULTS: The linear range of gallic acid, gallocatechin, corilagin, chebulagic acid and ellagic acid were 1.994-31.90, 0.159 8-2.556, 4.533-45.33, 14.75-88.49, 2.956-47.30 μg/mL (r≥0.999 8), respectively. The limits of detection were 0.025 6, 0.027 1, 0.052 9, 0.186 7, 0.133 1 μg/mL; the limits of quantitation were 0.085 1, 0.089 3, 0.170 6, 0.615 2, 0.441 9 μg/mL, respectively. RSDs of precision, stability (24 h) and reproducibility tests were all<2.0% (n=6 or n=7). The average recoveries were 96.19%-101.49% (RSD=0.98%-1.41%, n=6). 12 batches of samples had 3 main components. They could be divided into 2 categories. P. emblica from Rong county of Yulin city in Guangxi Zhuang Autonomous Region had highest comprehensive principal component score and best quality. CONCLUSIONS: Established method is simple, accurate and reliable, and can be used for the simultaneous determination of gallic acid, gallocatechin, corilagin, chebulagic acid and ellagic acid in P. emblica. The quality of P. emblica from different producing areas in Guangxi is different. |
