TITLE:
摘要: 目的:建立一种用于痱子粉中樟脑、薄荷脑含量测定的全封闭、免处理的定量分析方法。方法:采用顶空气相色谱法,以正辛醇为内标,以1-乙基-3-甲基咪唑四氟硼酸盐为顶空溶剂。色谱柱为PEG-20M毛细管柱,检测器为氢火焰离子化检测器,平衡温度为140 ℃,平衡时间为30 min,柱温为160 ℃,进样口温度为170 ℃,检测器温度为200 ℃,载气为氮气,流速为41 ml/min,分流比为18 ∶ 1,顶空进样体积为1 ml。结果:樟脑、薄荷脑检测质量浓度线性范围分别为5.15×10-3~515、5.40×10-3~540 μg/ml(r=0.998 4、0.998 2);精密度、稳定性、重复性试验的RSD<2%;回收率分别为99.0%~101.3%、98.3%~101.9%,RSD分别为0.9%、1.2%(n=9)。结论:该方法简单、准确、灵敏、重复性好,适用于痱子粉中樟脑、薄荷脑的含量测定,为固体药物制剂中半挥发性成分的定量分析提供了一个准确、可靠的分析手段。
ABSTRACT: OBJECTIVE:To establish a fully enclosed and free treatment quantitative analysis method for the contents determination of camphor and menthol in Prickly heat powder. METHODS: GC chromatography was conducted with internal standard of N-butanol and headspace solvent of 1-ethyl-3-methylimidazolium tetrafluoroborate. The column was PEG-20M capillary column, detector was hydrogen flame ionization detector, equilibrium temperature was 140 ℃, equilibrium time was 30 min, the column temperature was 160 ℃, the inlet temperature was 170 ℃, the detector temperature was 200 ℃, carrier gas was nitrogen, flow rate was 41 ml/min, split ratio was 18 ∶ 1, and the headspace volume was 1 ml. RESULTS: The linear range was 5.15×10-3-515 μg/ml(r=0.998 4) both for camphor and 5.40×10-3-540 μg/ml(r=0.998 2) for menthol; RSDs of precision, stability and reproducibility tests were lower than 2%; recoveries were 99.0%-101.3%(RSD=0.9%,n=9) and 98.3%-101.9%(RSD=1.2%,n=9), respectively. CONCLUSIONS: This method is simple, accurate, sensitive and reproducible, and can be used for the contents determination of camphor and menthol in Prickly heat powder, which provides an accurate and reliable analysis method for the quantitative analysis of semi-volatile components in solid pharmaceutical preparations.
