TITLE:
摘要: 目的:比较野生与栽培黄管秦艽中马钱苷酸、獐牙菜苦苷、6′ -O-β-D-葡萄糖基龙胆苦苷、龙胆苦苷、獐牙菜苷、异荭草苷和异牡荆苷7种指标成分的含量,为黄管秦艽的合理使用提供依据。方法:采用超高效液相色谱法。色谱柱为ACQUITY UPLC® BEH C18(50 mm×2.1 mm,1.7 μm),流动相为甲醇-0.04%磷酸溶液(梯度洗脱),流速为0.3 ml/min,检测波长为242 nm,柱温为30 ℃。结果:马钱苷酸、獐牙菜苦苷、6′ -O-β-D-葡萄糖基龙胆苦苷、龙胆苦苷、獐牙菜苷、异荭草苷和异牡荆苷进样量分别在2.1~537.1、1.05~270、0.92~236、11.1~2 830、0.75~192、0.167~102、0.216~52.80 μg范围内与峰面积积分值线性关系良好(r≥0.999 5),平均回收率为97.72%~99.84%(RSD≤3.39%,n=6)。野生黄管秦艽中马钱苷酸、獐牙菜苦苷、6′ -O-β-D-葡萄糖基龙胆苦苷、龙胆苦苷、獐牙菜苷、异荭草苷的含量略高于栽培黄管秦艽,而异牡荆苷含量略低于栽培黄管秦艽,但差异均无统计学意义(P>0.05)。野生与栽培品中龙胆苦苷与马钱苷酸含量总和均高于2.5%,达到2015年版《中国药典》(一部)要求。结论:野生与栽培黄管秦艽7种指标成分的含量差异无统计学意义,且药典规定成分的含量都合格。
ABSTRACT: OBJECTIVE: To compare the contents of loganic acid, swertiamarin, 6′ -O-β-D-glucosyl gentiopicroside, gentiopicroside, sweroside, isoorientin and isovitexin in wild and cultivated Gentiana officinalis, and to provide basis for rational use of G. officinalis. METHODS: UPLC method was adopted. The separation was performed on ACQUITY UPLC® BEH C18 column (50 mm×2.1 mm, 1.7 μm) with mobile phase consisted of methanol-0.04% phosphoric acid (gradient elution) at the flow rate of 0.3 ml/min. The detection wavelength was set at 242 nm, and column temperature was 30 ℃. RESULTS: For loganic acid, swertiamarin, 6′ -O-β- D-glucosyl gentiopicroside, gentiopicroside, sweroside, isoorientin and isovitexin, a good linearity was obtained in the range of 2.1-537.1 μg, 1.05-270 μg, 0.92-236 μg, 11.1-2 830 μg, 0.75-192 μg, 0.167-102 μg, 0.216-52.80 μg (r≥0.999 5), respectively. Their average recoveries were 97.72%-99.84% (RSD≤3.39%, n=6). The contents of loganic acid, swertiamarin, 6′ -O-β-D-glucosyl gentiopicroside, gentiopicroside, sweroside and isoorientin in the wild sample were higher than in the cultivated; the content of isovitexin was lower than the cultivated, but there was no statistical significance (P>0.05). The sum of gentiopicroside and loganin acid content were all higher than 2.5% in both wild and cultivated samples, and met the requirements of 2015 edition of Chinese Pharmacopoeia (first part). CONCLUSIONS: The content difference of 7 indicative constituents in wild and cultivated G. officinalis is not statistically significant, and the indicative constituents of the pharmacopoeia is qualified.
