TITLE:
摘要: 目的:建立测定左卡尼汀注射液中有关物质2(5H)-呋喃酮含量的方法。方法:采用高效液相色谱法。色谱柱为APS-2 Hypersil,流动相为0.05 mol/L磷酸盐缓冲液(pH4.7)-乙腈(35 ∶ 65,V/V),流速为1.0 ml/min,检测波长为205 nm,柱温为30 ℃,进样量为20 μl。结果:2(5H)-呋喃酮检测质量浓度线性范围为0.001 974~0.987 mg/ml(r=0.999 9);定量限为4.0 ng,检测限为1.2 ng;精密度、稳定性、重复性试验的RSD<2%;加样回收率为97.04%~103.36%(RSD=0.24%,n=9)。结论:该方法简便、灵敏度高、重复性好,适用于左卡尼汀注射液中有关物质2(5H)-呋喃酮的含量测定。
ABSTRACT: OBJECTIVE: To establish a method for the determination of 2(5H)-Furanone in Levocanitine injection. METHODS: HPLC was performed on the column of APS-2 HYPERSIL with mobile phase of 0.05 mol/L potassium dihydrogen phosphate buffer solution (pH4.7)-acetonitrile (35 ∶ 65,V/V) at a flow rate of 1.0 ml/min, the detection wavelength was 205 nm, column temperature was 30 ℃, and injection volume was 20 μl. RESULTS: The linear range of 2(5H)-Furanone was 0.001 974-0.987 mg/ml; the limit of quantitation was 4.0 ng, the limit of detection was 1.2 ng; RSDs of precision, stability and reproducibility tests were lower than 2%; recovery was 97.04%-103.36%(RSD=0.24%,n=9). CONCLUSIONS: The method is simple, accurate, sensitive and reproducible, and suitable for the content determination of 2(5H)-Furanone in Levocanitine injection.
