摘要: 目的:观察温胃调肠颗粒对便秘型肠易激综合征(IBS-C)模型大鼠的改善作用,并探讨其可能的作用机制。方法:将60只SD大鼠随机分为正常组、模型组、莫沙必利组(阳性药物,50 mg/kg)和温胃调肠颗粒低、中、高剂量组(3、6、12 g/kg),每组10只。除正常组外,其余各组大鼠ig冰水混合物复制IBS-C模型。造模后,各给药组大鼠ig相应药物,正常组和模型组大鼠ig等体积生理盐水,每天1次,连续14 d。给药结束后,测定大鼠24 h排便粒数、粪便含水率;采用酶联免疫吸附法测定大鼠血清中5-羟色胺(5-HT)和降钙素基因相关肽(CGRP)的含量;分别采用实时荧光定量聚合酶链式反应法和Western  blot法测定大鼠结肠组织中5-羟色胺转运蛋白(SERT)、CGRP mRNA及其蛋白表达水平。结果:与正常组比较,模型组大鼠24 h排便粒数、粪便含水率减少,血清中5-HT和CGRP含量增加,结肠组织中SERT和CGRP mRNA及其蛋白水平升高(P<0.05或P<0.01);与模型组比较,各给药组大鼠上述指标均得到不同程度改善,除温胃调肠颗粒低剂量组大鼠结肠组织SERT mRNA降低不显著外,其余各组差异均有统计学意义(P<0.05或P<0.01)。结论:温胃调肠颗粒可促进IBS-C模型大鼠的排便,改善便秘症状;其机制可能与降低血清和肠组织中5-HT、CGRP等神经递质的水平有关。
ABSTRACT: OBJECTIVE: To observe the improvement effects of Wenwei tiaochang granule on constipation-predominant irritable bowel syndrome (IBS-C) model rats, and to discuss its mechanism. METHODS: 60 SD rats were randomly divided into normal group, model group, mosapride group (positive drug, 50 mg/kg) and Wenwei tiaochang granule low-dose, medium-dose and high-dose groups (3, 6, 12 g/kg), with 10 rats in each group. Except for normal group, other groups were given ice water mixture intragastrically to induce IBS-C model. After modeling, treatment groups were given relevant medicine intragastrically, and normal group and model group were given constant volume of normal saline intragastrically, once a day, for consecutive 14 d. After medication, 24 h dejection amounts and water rate of manure were measured. The serum contents of 5-HT and CGRP were determined by ELISA. RT-PCR and Western blot assay were used to determine mRNA and protein expression of SERT and CGRP in colon tissue. RESULTS: Compared with normal group, 24 h dejection amounts and water rate of manure were decreased in model group, serum contents of 5-HT and CGRP were increased, and mRNA and protein expression of SERT and CGRP in colon tissue were also increased (P<0.05 or P<0.01). Compared with model group, above indexes of treatment groups were all improved to different extent, with statistical significance (P<0.05 or P<0.01), except the decrease of mRNA expression of SERT in colon tissue was not significant in Wenwei tiaochang granule low-dose group. CONCLUSIONS: Wenwei tiaochang granule can improve the dejection and symptoms of constipation in IBS-C model rats, which maybe associated with decreasing the levels of 5-HT, CGRP and other neurotransmitter in serum and intestinal tissue.
